There are lots of nice roads in that area peteski9 if you don't mind some undulations here & there. I'm not sure of the roads to the West or South but have ridden to both Castlemaine and Trentham from Daylesford recently. A nice 100kish loop would be to travel North to Castlemaine on the Midland Hwy. At Castlemaine turn West to Newstead and then at Newstead return to Daylesford via the Daylesford/Newstead road. For added hill thrills take a left at the Hepburn/Newstead road to continue through Shepherds flat. Using this return route instead of the main road takes you up through Hepburn Springs and there are some steepish climbs.(There is also a friendly pub half way.
The Midland Hwy can be busy but there is a shoulder if you need it. It is also slightly downhill (travelling North) so if you are lucky enough to have a tailwind you can sit on quite a high speed for the 30 odd k's.
Another nice ride would be East over to Trentham. There is a nice quite road called Coliban road about 3km's before you get to Trentham. That takes you through part of the Wombat State Forest to Lauriston. You could return to Daylesford via the Malmsbury/Daylesford road which is nice.
That part of the world had heaps of good bicycle rides. Maybe have a look at bikely
and have a search for Daylesford.
